Vignola and della Porta eliminated traditional side aisles, directing the congregation into a single wide nave under a cross vault. The layout prioritized acoustics, ensuring sermons carried without echoes.

Della Porta revised the exterior into the first recognized Baroque facade, stacking vertical elements over Vignola's flatter scheme. Inside, identical chapels behind gated balustrades replace aisles, while polychrome marble, gilding, and vaulted frescoes fill the space.

Michelangelo originally offered to design the structure without payment out of personal devotion, though patronage ultimately went to Farnese.
