The institution operates as the primary Jesuit university in Rome, educating clergy and scholars from across the globe.

Ignatius of Loyola established the original school near Capitoline Hill with financial backing from Francis Borgia. Pope Paul authorized the institution to confer advanced degrees in theology and philosophy. Substantial expansion under Pope Gregory produced a dedicated complex designed by Bartolomeo Ammannati at Piazza Collegio Romano, establishing the Gregorian name and housing scholarship in mathematics and astronomy.

Scholarly work here shaped scientific history, including the development of the Gregorian calendar by Christopher Clavius and mathematical research by Athanasius Kircher. The adjoining Church of Sant'Ignazio originated as the university chapel. Following national unification, government authorities expropriated the central property for a public school, which compelled the administration to transfer operations to Quirinal Hill.
